Pencillin Sensitisation tests, Scratch Test
Sensitisation test: There are no reliable sensitisation tests which ca n infallibly predict the occurance of immediate type of reaction to a non protein drug nor is it safe to perform ID tests with suspected drugs as they may sometimes cause severe and even fatal anaphylaxis.
Potassium benzyl penicillin is an excellent antigenic substance which in an aquous dilution containing 10U units/ml is usually used for tests. The penieilloyl group which is the most important antigenic determinent, is a degradation product of penicillin. The penicilloyl polylysine (PPL) skin test if positive is accepted by many as a valid indication of penicillin allergy.
Scratch Test:- Put a drop of penicillin solution on the skin of forearm and through that make a scratch with a blunt scalpel. A wheal and flare develops on the scratch mark.
